Physiological response of the retinal pigmented epithelium to 3‐ns pulse laser application, in vitro and in vivo

Background To treat healthy retinal pigmented epithelium (RPE) with the 3-ns rejuvenation therapy (2RT) laser and to investigate subsequent wound-healing response of these cells. Methods Primary rat RPE cells were treated 2RT at a range energy settings. Treated fixed up 7 days post-irradiation assessed for expression proteins associated wound-healing. For in vivo treatments, eyes Dark Agouti rats exposed tissues collected post-irradiation. Isolated wholemount preparations examined structural protein changes. Results Cultured ablated by an energy-dependent manner. In all cases, cell layer repopulated completely within days. Replenishment was heat shock protein, Hsp27, intermediate filament proteins, vimentin nestin, cycle-associated cyclin D1. Cellular tight junctions lost lased regions but re-expressed when replenishment complete. vivo, treatment gave rise both localised denudation repopulation lesion sites. Cell increased D1, Hsp27 αB-crystallin. Conclusions The able target vitro causing debridement consequent stimulation leading reformation.
